In industrial applications of laser welding of aluminum alloys, it is often a challenge to find optimal process parameters to produce welded joints of good quality. This study investigates the effect of changes in welding parameters on the resulting welding defects and tensile properties of laser-welded aluminum alloy joints. A high-power disk laser was used for welding 2.54 mm thick AA6061-T6 sheets with four different sets of process parameters. X-ray computed microtomography and full-field microhardness mapping were first used to characterize the as-welded joints. Surface strain mapping based on digital image correlation and a two-mirror optical setup was applied to tensile testing of welded joints up to ductile failure. It was found that the fusion zone of all four laser-welded joints has similar microhardness levels and spatial distributions. Small pores were detected in all four weld joints, except one had a few large pores. Both microstructural heterogeneities and surface geometric irregularities were found to induce highly non-uniform local tensile deformation in laser-welded joints. One set of laser welding parameters was identified to produce the aluminum welds with the best tensile properties even though its weld joint may contain a few relatively large pores. The study was conducted to quantify laryngeal cartilage matrix composition and to investigate its relationship with cartilage shape in a mouse model. A sample of 30 mice (CD‐1 mouse, Mus musculus) from five age groups (postnatal Days 2, 21, 90, 365, and 720) were used. Three‐dimensional mouse laryngeal thyroid cartilage reconstructions were generated from contrast‐enhanced micro‐computed tomography (CT) image stacks. Cartilage matrix composition was estimated as Hounsfield units (HU). HU were determined by overlaying 3D reconstructions as masks on micro‐CT image stacks and then measuring the attenuation. Cartilage shape was quantified with landmarks placed on the surface of the thyroid cartilage. Shape differences between the five age groups were analyzed using geometric morphometrics and multiparametric analysis of landmarks. The relationship between HU and shape was investigated with correlational analyses. Among five age groups, HU became higher in older animals. The shape of the thyroid cartilage changes with age throughout the entire life of a mouse. The changes in shape were not synchronized with changes in cartilage matrix composition. The thyroid cartilage of young and old M. musculus larynx showed a homogenous mineralization pattern. Objectives: Reconstruction of fossil hominin manual behaviors often relies on comparative analyses of extant hominid hands to understand the relationship between hand use and skeletal morphology. In this context, the intermediate phalanges remain understudied. Thus, here we investigate cortical bone morphology of the intermediate phalanges of extant hominids and compare it to the cortical structure of the proximal phalanges, to investigate the relationship between cortical bone structure and inferred loading during manual behaviors. Materials and Methods: Using micro‐CT data, we analyze cortical bone structure of the intermediate phalangeal shaft of digits 2–5 in Pongo pygmaeus (n = 6 individuals), Gorilla gorilla (n = 22), Pan spp. (n = 23), and Homo sapiens (n = 23). The R package morphomap is used to study cortical bone distribution, cortical thickness and cross‐sectional properties within and across taxa. Results: Non‐human great apes generally have thick cortical bone on the palmar shaft, with Pongo only having thick cortex on the peaks of the flexor sheath ridges, while African apes have thick cortex along the entire flexor sheath ridge and proximal to the trochlea. Humans are distinct in having thicker dorsal shaft cortex as well as thick cortex at the disto‐palmar region of the shaft. Discussion: Variation in cortical bone distribution and properties of the intermediate phalanges is consistent with differences in locomotor and manipulative behaviors in extant great apes. Comparisons between the intermediate and proximal phalanges reveals similar patterns of cortical bone distribution within each taxon but with potentially greater load experienced by the proximal phalanges, even in knuckle‐walking African apes. Objectives: Estimating the sex of cremated human remains is difficult. The petrous bone frequently survives the cremation due to its density. Wahl observed the lateral angle to be sexually dimorphic in the 1980s. Previous studies showed various cut‐off points to separate females from males, which are hardly replicable and difficult to apply. We want to test the Wahl method and compare it to a new landmark‐based version. Materials and Methods: In this study, we measured the lateral angle of 35 cremated petrous bones from late bronze age Austria using micro‐CT scans. Technical errors of measurement were calculated for two different methods to intersect the internal acoustic meatus virtually in the midline (manual or landmark‐based intersection). Furthermore, sex was estimated based on morphological features and metric measurements. This information was used in logistic regression modeling to define a cut‐off point in our sample. Results: The technical errors of measurement suggested that a landmark‐based method was more precise in comparison to a manual intersection which was much more intuitive. Inter‐ and intra‐observer errors were low which improved reliability. The logistic regression model produced good results in our sample (p = 0.02, R2 = 0.38, accuracy = 0.8). The mean lateral angle was similar to studies which focused on prehistoric cremated petrous bones. Discussion: The proposed landmark‐based method was precise, quick, and could be easily applied, even by unexperienced researchers. The size of the lateral angle seemed to be population‐specific but also dependent on the method applied. We recommend to use the proposed landmark‐based method which is more precise. BACKGROUND: The lower cervical vertebral pedicle is the main stress site of the posterior column of the spine, which is of great significance for the maintenance of the stability of the human center of gravity and the reduction of shock. At present, there are few reports on the characteristics of the internal bone trabeculae, and the characteristics of the joint site of the vertebral pedicle with the articular process and the vertebral body. It is urgent to understand the fine anatomical structure of the vertebral pedicle and the relationship and function of each part. OBJECTIVE: To observe the microanatomical morphology of the vertebral pedicle by Micro-CT scanning of cervical vertebra specimens, and to measure and analyze the microstructure and morphometric parameters of the bone trabecula in the cervical pedicle under normal conditions to evaluate the safety performance of the cervical spine. METHODS: Micro-CT scanning was performed on 31 sets of cervical vertebrae C3-C7. By checking and reconstructing the areas of interest in the bone trabecular within the vertebral pedicle, the morphological characteristics and distribution direction of the bone trabecular within the cervical pedicle were observed, and the bone microstructure parameters were detected, and the differences in the bone microstructure of the C3-C7 vertebral pedicle were analyzed and compared. RESULTS AND CONCLUSION: (1) The Micro-CT images showed that the honeycomb bone trabeculae of the pedicle of the lower cervical spine presented a complex network of microstructures. The trabeculae near the cortical bone were lamellar and relatively compact, extending forward toward the vertebral body and backward toward the articular process lamina. Abatoid bone trabeculae extended into the medullary cavity and transformed into a network structure, and then into rod-shaped bone trabeculae. The rod-shaped bone trabeculae were sparsely distributed in the medullary cavity. (2) Statistical results of morphological parameters of bone trabeculae showed that bone volume fraction values in C4 and C5 were higher than that in C7 (P < 0.05). The bone surface/ bone volume value in C7 was higher than that in C3, C4 and C6 (P < 0.05). The bone surface density of bone trabeculae in C7 was higher than that in C3, C4, C5 and C6 (P < 0.05). Trabecular thickness in C7 was higher than that in C3, C4 and C5 (P < 0.05). Bone surface/bone volume and bone surface density of the left pedicle bone trabecular were greater than those on the right side (P < 0.05). (3) The microstructural changes of C3-C7 were summarized, in which the load capacity and stress of the C7 pedicle were poor, and the risk of injury was high in this area. [ABSTRACT FROM AUTHOR]

2.5D woven structures are of interest in new thermal protection systems for their excellent interlayer properties and structural stability. In this paper, dense (CDP) and lightweight (CLP) carbon/phenolic 2.5D woven composites were prepared. Quasi‐static tensile experiments were performed on the specimens before ablation (TCDP‐0, TCLP‐0) and after ablation for 30 s (TCDP‐30, TCLP‐30). A quarter macro–meso hybrid model that takes into account the degradation of material properties after ablation and ablative crack pores was innovatively proposed. The predicted tensile elastic modulus is within 9% error from the experimental results. The results show that TCDP‐30 and TCLP‐30 have rapid damage expansion and lethal injury at smaller displacements than the pre‐ablation specimens. The displacement of the maximum load point of TCDP‐30 is 38.7% lower than that of TCLP‐30. The forms of damage in TCDP‐30 are dominated by matrix fragmentation and matrix cracks. The damage forms of TCLP‐30 are dominated by debonding and delamination. Monoculture switchgrass and restored prairie are promising perennial feedstock sources for bioenergy production on the lands unsuitable for conventional agriculture. Such lands often display contrasting topography that influences soil characteristics and interactions between plant growth and soil C gains. This study aimed at elucidating the influences of topography and plant systems on the fate of C originated from switchgrass plants and on its relationships with soil pore characteristics. For that, switchgrass plants were grown in intact soil cores collected from two contrasting topographies, namely steep slopes and topographical depressions, in the fields in multi-year monoculture switchgrass and restored prairie vegetation. The 13C pulse labeling allowed tracing the C of switchgrass origin, which X-ray computed micro-tomography enabled in-detail characterization of soil pore structure. In eroded slopes, the differences between the monoculture switchgrass and prairie in terms of total and microbial biomass C were greater than those in topographical depressions. While new switchgrass increased the CO2 emission in depressions, it did not significantly affect the CO2 emission in slopes. Pores of 18–90 µm Ø facilitated the accumulation of new C in soil, while > 150 µm Ø pores enhanced the mineralization of the new C. These findings suggest that polyculture prairie located in slopes can be particularly beneficial in facilitating soil C accrual and reduce C losses as CO2. [ABSTRACT FROM AUTHOR]

Calcium oxalate kidney stones, the most prevalent type of kidney stones, undergo a multi-step process of crystal nucleation, growth, aggregation, and secondary transition. The secondary transition has been rather overlooked, and thus, the effects on the disease and the underlying mechanism remain unclear. Here, we show, by periodic micro-CT images of human kidney stones in an ex vivo incubation experiment, that the growth of porous aggregates of calcium oxalate dihydrate (COD) crystals triggers the hardening of the kidney stones that causes difficulty in lithotripsy of kidney stone disease in the secondary transition. This hardening was caused by the internal nucleation and growth of precise calcium oxalate monohydrate (COM) crystals from isolated urine in which the calcium oxalate concentrations decreased by the growth of COD in closed grain boundaries of COD aggregate kidney stones. Reducing the calcium oxalate concentrations in urine is regarded as a typical approach for avoiding the recurrence. However, our results revealed that the decrease of the concentrations in closed microenvironments conversely promotes the transition of the COD aggregates into hard COM aggregates. Both bone morphogenetic protein 2 (BMP-2) and abaloparatide are used to promote bone formation. However, there is no consensus about their optimal administration. We investigated the optimal administration theory for the pairing of BMP-2 and abaloparatide in a rat spinal fusion model. Group I was only implanted in carriers and saline. Carriers with 3 µg of recombinant human BMP-2 (rhBMP-2) were implanted in other groups. Abaloparatide injections were administered three times a week for group III (for a total amount of 120 µg/kg in a week) and six times a week for group IV (for a total amount of 120 µg/kg in a week) after surgery. They were euthanized 8 weeks after the surgery, and we explanted their spines at that time. We assessed them using manual palpation tests, radiography, high-resolution micro-computed tomography (micro-CT), and histological analysis. We also analyzed serum bone metabolism markers. The fusion rate in Groups III and IV was higher than in Group I, referring to the manual palpation tests. Groups III and IV recorded greater radiographic scores than those in Groups I and II, too. Micro-CT analysis showed that Tbs. Sp in Groups III and IV was significantly lower than in Group I. Tb. N in Group IV was significantly higher than in Group I. Serum marker analysis showed that bone formation markers were higher in Groups III and IV than in Group I. On the other hand, bone resorption markers were lower in Group IV than in Group I. A histological analysis showed enhanced trabecular bone osteogenesis in Group IV. Frequent administration of abaloparatide may be suitable for the thickening of trabecular bone structure and the enhancement of osteogenesis in a rat spinal fusion model using BMP-2 in insufficient doses. Aims: The aim of this randomized controlled clinical trial was to compare the gene expression, micro‐CT, histomorphometrical analysis between biphasic calcium phosphate (BCP) of 70/30 ratio and deproteinized bovine bone mineral (DBBM) in sinus augmentation. Materials and Methods: Twenty‐four patients in need for sinus floor augmentation through lateral approach were randomized into BCP 70/30 ratio or DBBM. After at least 6 months of healing, a total of 24 bone specimens were collected from the entire height of the augmented bone at the area of implant placement and underwent micro‐CT, histomorphometric and gene expression analysis. The 12 bone specimens of BCP 70/30 ratio were equally allocated to micro‐CT and histologic analysis (test group, n = 6) and gene expression analysis (test group, n = 6). Similarly, the 12 bone specimens of DBBM were also allocated to micro‐CT and histologic analysis (control group, n = 6) and gene expression analysis (control group, n = 6). The newly formed bone, remaining graft materials and relative change in gene expression of four target genes were assessed. Results: The micro‐CT results showed no statistically significant difference in the ratio of bone volume to total volume (BV/TV ratio) for the two groups (BCP 41.51% vs. DBBM 40.97%) and the same was true for residual graft material to total volume (GV/TV ratio, BCP 9.97% vs. DBBM 14.41%). Similarly, no significant difference was shown in the histological analysis in terms of bone formation, (BCP 31.43% vs. DBBM was 30.09%) and residual graft area (DBBM 40.76% vs. BCP 45.06%). With regards to gene expression, the level of ALP was lower in both groups of bone grafted specimens compared with the native bone. On the contrary, the level of OSX, IL‐1B and TRAP was higher in augmented bone of both groups compared with the native bone. However, the relative difference in all gene expressions between BCP and DBBM group was not significant. Conclusions: The BCP, HA/β‐TCP ratio of 70/30 presented similar histological and micro‐CT outcomes in terms of new bone formation and residual graft particles with DBBM. This article explores the potential therapeutic benefits of naringin, a compound found in citrus fruits, for individuals with Alzheimer's disease (AD). The study used an animal model of AD to investigate the effects of naringin on memory impairment, the volume of a specific area in the brain called the CA1 area, and neuronal loss. The results indicated that naringin improved memory, increased the volume of the CA1 area, and increased the number and density of neurons in the hippocampus. Ankylosing spondylitis (AS) is a chronic inflammatory disease, characterized by excessive new bone formation. We previously reported that the complement factor H-related protein-5 (CFHR5), a member of the human factor H protein family, is significantly elevated in patients with AS compared to other rheumatic diseases. However, the pathophysiological mechanism underlying new bone formation by CFHR5 is not fully understood. In this study, we revealed that CFHR5 and proinflammatory cytokines (TNF, IL-6, IL-17A, and IL-23) were elevated in the AS group compared to the HC group. Correlation analysis revealed that CFHR5 levels were not significantly associated with proinflammatory cytokines, while CFHR5 levels in AS were only positively correlated with the high CRP group. Notably, treatment with soluble CFHR5 has no effect on clinical arthritis scores and thickness at hind paw in curdlan-injected SKG, but significantly increased the ectopic bone formation at the calcaneus and tibia bones of the ankle as revealed by micro-CT image and quantification. Basal CFHR5 expression was upregulated in AS-osteoprogenitors compared to control cells. Also, treatment with CFHR5 remarkedly induced bone mineralization status of AS-osteoprogenitors during osteogenic differentiation accompanied by MMP13 expression. We provide the first evidence demonstrating that CFHR5 can exacerbate the pathological bone formation of AS. Therapeutic modulation of CFHR5 could be promising for future treatment of AS. The migration of gases is crucial to ensure the long-term feasibility of argillaceous formations for the deep disposal of radioactive waste. This paper presents an experimental investigation with a multi-scale perspective on the response to gas transport of initially saturated Boom Clay (Belgium). Gas injection tests have been performed under oedometer conditions at different controlled-volume rates, constant total vertical stress and different sample orientations (flow orthogonal or parallel to bedding planes). The results confirm soil expansion and consequent degradation during injection that has a significant impact on the aperture of localised gas pathways (fissures) and increases intrinsic permeability during the gas pressure dissipation stage. The analyses with complementary techniques (mercury intrusion porosimetry, field-emission scanning electron microscopy and X-ray micro-tomography) confirm the opening of fissures with different apertures and separations at the microstructural scale. Large-aperture fissures develop along the weaker bedding planes. These techniques allow the volume of fissures to be quantified, which does not significantly depend on gas flow direction, as also measured in the isotropic response of the gas effective permeability. A scalar damage variable derived from the fissured fraction has been used to assess the gas-entry pressure reduction and the intrinsic permeability increase after the gas tests in both directions. A wooden polychrome statuette of Virgin and Child in Majesty, currently in the Museum Montanelli in Prague, is actually a torso. It was created in the 11th or 12th century at the latest, being the oldest wooden polychrome statuette in the Czech Republic. Current research combining advanced imaging and micro‐analytical methods substantially contributed to the determination of its regional origin and detection and dating of later interventions. Identified pigments, including azurite, vivianite and smalt, together with changes in gilding technology, were found to be essential in tracing the story of the statuette over time. The purpose of this study was to provide detailed knowledge of the morphological properties of ash particles, including the volumetric fractions and 3D distributions of phosphates that lay within them. The ash particles came from digested sewage sludge co-combusted with K- and Si-rich wheat straw or K-rich sunflower husks. X-ray micro-tomography were combined with elemental composition and crystalline phase information to analyse the ash particles in 3D. Analyses of differences in the X-ray attenuation enabled calculation of 3D phosphate distributions that showed high heterogeneity in the slag particles. This is underscored by a distinct absence of phosphates in iron-rich and silicon-rich parts. The slag from silicate-based wheat straw mixtures had lower average attenuation than that from sunflower husks mixtures, which contained more calcium. Calculated shares of phosphates between 7 and 17 vol% were obtained, where the highest value for a single assigned phosphate was observed in hard slag from wheat straw with 10 % sewage sludge. The porosity was notably higher for particles from pure wheat straw combustion (62 vol%), compared to the other samples (15–35 vol%). A high open pore volume fraction (60–97 vol%) indicates that a large part of the pores can be accessed by the surroundings. For all samples, more than 60 % of the discrete (closed) pores had an equivalent diameter < 30 μm, while the largest volume fraction consisted of pores with an equivalent diameter > 75 μm. Slag from sunflower husk mixtures had larger pore volumes and a greater relative number of discrete pores >75 µm compared to wheat straw mixtures. [ABSTRACT FROM AUTHOR]

Attempts to explain the origin and diversification of vertebrates have commonly invoked the evolution of feeding ecology, contrasting the passive suspension feeding of invertebrate chordates and larval lampreys with active predation in living jawed vertebrates. Of the extinct jawless vertebrates that phylogenetically intercalate these living groups, the feeding apparatus is well-preserved only in the early diverging stem-gnathostome heterostracans. However, its anatomy remains poorly understood. Here, we use X-ray microtomography to characterize the feeding apparatus of the pteraspid heterostracan Rhinopteraspis dunensis (Roemer, 1855). The apparatus is composed of 13 plates arranged approximately bilaterally, most of which articulate from the postoral plate. Our reconstruction shows that the oral plates were capable of rotating around the transverse axis, but likely with limited movement. It also suggests the nasohypophyseal organs opened internally, into the pharynx. The functional morphology of the apparatus in Rhinopteraspis precludes all proposed interpretations of feeding except for suspension/deposit feeding and we interpret the apparatus as having served primarily to moderate the oral gape. This is consistent with evidence that at least some early jawless gnathostomes were suspension feeders and runs contrary to macroecological scenarios that envisage early vertebrate evolution as characterized by a directional trend towards increasingly active food acquisition. Cone-beam computed tomography (CBCT) is widely used in dental imaging, small animal imaging, radiotherapy, and non-destructive industrial inspection. The quality of CBCT images depends on the precise knowledge of the CBCT system's alignment. We introduce a distinct procedure, 'precision alignment loop (PAL)', to calibrate any CBCT system with a circular trajectory. We describe the calibration procedure by using a line-beads phantom, and how PAL determines the misalignments from a CBCT system. PAL also yields the uncertainties in the simulated calibration to give an estimate of the errors in the misalignments. From the analytical simulations, PAL can precisely obtain the source-to-rotation axis distance, and the geometric center G, 'the point in z -axis meets the detector', where the z -axis is coincident with the line from the x-ray source that intersects the axis of the rotation orthogonally. The uncertainties of three misalignment angles of the detector are within ±0.05°, which is close to ±0.04° for the results of Yang et al (2017 Med. Phys. 44 1692), but our method is easy and simple to implement. Our distinct procedure, on the other hand, yields the calibration of a micro-CT system and an example of reconstructed images, showing our calibration method for the CBCT system to be simple, precise, and accurate. Global demand for underground gas storage (UGS) is steadily increasing, with the limestone-based UGS system situated in the Sichuan Basin of China gathering considerable interest in recent years. However, studies focusing on the fundamental mechanisms of the injection-production process in these systems are limited. Moreover, existing studies utilizing physical experimental methods frequently fall short in effectively visualizing micro-flow or incorporating real core samples from the reservoir. To address these gaps, we performed a coreflood experiment, integrating micro-Computed Tomography (CT) scanning to investigate mechanisms of fluid flow and storage capacity during the injection and production cycles in limestone reservoirs. Our approach involved utilizing core plugs with artificially engraved fracture-vuggy structures, which mimic the characteristics of the reservoir. Micro-CT scans were performed to visualize the microscopic changes in fractured-vuggy structures and the distribution of irreducible water during each cycle. This study reveals that increased cycles correspondingly affect gas storage capacity, particularly by expanding it in relative larger vuggy structures while reducing it in finer fissure network structures. The amount of irreducible water decreases after injection-production cycles, likely being expelled alongside the extracted dry gas. This plays a critical role in expanding the storage capacity in larger vuggy systems. Conversely, there is a decrease in storage capacity within fissure network systems, as the irreducible water is replaced by gas. This leads to a reduction in the opening force of the fine conduit. The dense matrix has a very limited effect on the flow mechanism and its influence on storage capacity. Overall, these findings offer practical insights for optimizing injection and production strategies in limestone UGS systems within the Sichuan Basin, contributing to a deeper understanding and efficient utilization of this vital infrastructure. Detecting breast tissue alterations is essential for cancer diagnosis. However, inherent bidimensionality limits histological procedures' effectiveness in identifying these changes. Our study applies a 3D virtual histology method based on X-ray phase-contrast microtomography (PhC μ CT), performed at a synchrotron facility, to investigate breast tissue samples including different types of lesions, namely intraductal papilloma, micropapillary intracystic carcinoma, and invasive lobular carcinoma. One-to-one comparisons of X-ray and histological images explore the clinical potential of 3D X-ray virtual histology. Results show that PhC μ CT technique provides high spatial resolution and soft tissue sensitivity, while being non-destructive, not requiring a dedicated sample processing and being compatible with conventional histology. PhC μ CT can enhance the visualization of morphological characteristics such as stromal tissue, fibrovascular core, terminal duct lobular unit, stromal/epithelium interface, basement membrane, and adipocytes. Despite not reaching the (sub) cellular level, the three-dimensionality of PhC μ CT images allows to depict in-depth alterations of the breast tissues, potentially revealing pathologically relevant details missed by a single histological section. Compared to serial sectioning, PhC μ CT allows the virtual investigation of the sample volume along any orientation, possibly guiding the pathologist in the choice of the most suitable cutting plane. Background: Imbalance in energy regulation is a major cause of insulin resistance and diabetes. Melanocortin-4 receptor (MC4R) signaling at specific sites in the central nervous system has synergistic but non-overlapping functions. However, the mechanism by which MC4R in the arcuate nucleus (ARC) region regulates energy balance and insulin resistance remains unclear. Methods: The MC4Rflox/flox mice with proopiomelanocortin (POMC) -Cre mice were crossed to generate the POMC-MC4Rflox/+ mice. Then POMC-MC4Rflox/+ mice were further mated with MC4Rflox/flox mice to generate the POMC-MC4Rflox/flox mice in which MC4R is selectively deleted in POMC neurons. Bilateral injections of 200 nl of AAV-sh-Kir2.1 (AAV-sh-NC was used as control) were made into the ARC of the hypothalamus. Oxygen consumption, carbon dioxide production, respiratory exchange ratio and energy expenditure were measured by using the CLAMS; Total, visceral and subcutaneous fat was analyzed using micro-CT. Co-immunoprecipitation assays (Co-IP) were used to analyze the interaction between MC4R and Kir2.1 in GT1-7 cells. Results: POMC neuron-specific ablation of MC4R in the ARC region promoted food intake, impaired energy expenditure, leading to increased weight gain and impaired systemic glucose homeostasis. Additionally, MC4R ablation reduced the activation of POMC neuron, and is not tissue-specific for peripheral regulation, suggesting the importance of its central regulation. Mechanistically, sequencing analysis and Co-IP assay demonstrated a direct interaction of MC4R with Kir2.1. Knockdown of Kir2.1 in POMC neuron-specific ablation of MC4R restored the effect of MC4R ablation on energy expenditure and systemic glucose homeostasis, indicating by reduced body weight and ameliorated insulin resistance. Conclusion: Hypothalamic POMC neuron-specific knockout of MC4R affects energy balance and insulin sensitivity by regulating Kir2.1. Motion is the basis of nearly all animal behavior. Evolution has led to some extraordinary specializations of propulsion mechanisms among invertebrates, including the mandibles of the dracula ant and the claw of the pistol shrimp. In contrast, vertebrate skeletal movement is considered to be limited by the speed of muscle, saturating around 250 Hz. Here, we describe the unique propulsion mechanism by which Danionella cerebrum, a miniature cyprinid fish of only 12 mm length, produces high amplitude sounds exceeding 140 dB (re. 1 µPa, at a distance of one body length). Using a combination of high-speed video, micro-computed tomography (micro-CT), RNA profiling, and finite difference simulations, we found that D. cerebrum employ a unique sound production mechanism that involves a drumming cartilage, a specialized rib, and a dedicated muscle adapted for low fatigue. This apparatus accelerates the drumming cartilage at over 2,000 g, shooting it at the swim bladder to generate a rapid, loud pulse. These pulses are chained together to make calls with either bilaterally alternating or unilateral muscle contractions. In this article, we investigate acid fracturing, a promising method to increase the effectiveness of fractures in high temperature carbonate reservoirs. In order to determine the performance of acid fracturing in carbonate reservoirs, we conducted a lab scaled true triaxial acid fracturing experiment on high temperature carbonate rock. The fracture initiation, propagation behavior, and conductivity under the action of acid solution were analyzed. The 3-D micro computed tomography was applied to illuminate the fracture network patterns; coefficient of stress difference, acid etching time, and acid concentration were also analyzed. Background: Gulab jamun is a delicious dairy food items popular not only the Indian subcontinent but also in all over the world. Now a days, people are health conscious towards the healthy diet having low in fat content. However, Vacuum frying (VF) is one of the best technologies which produce high quality food items having low fat content and better appearance with respect to atmospheric frying (AF). Methods: For the preparation of gulab jamun, the dough balls were fried under the VF (oil temperature: 125, 135, 145C, vacuum pressure: 15, 20, 25 cm Hg and time: 8, 10, 12 min) and AF (temperature: 160C, time: 4 min) conditions. The prepared gulab jamun was compared with different parameters such as moisture content, oil content, color, volume expansion, texture and structure. Result: The optimized gulab jamun was obtained using response surface methodology at temperature 137.82C, pressure 25 cm Hg and time 9.38 min. The product had less in oil content, intense in color, smooth and soft texture (texture analyzer) and porous in structure (X-ray micro-CT scan) as compared to atmospheric fried gulab jamun. This study evaluates various methods for estimating soil permeability using microtomography-derived data and compares them to the conventional laboratory approaches. Different methods, including measurement in custom-designed permeameter at micro-CT-compatible scale, empirical equations, simulated sifting, semi-theoretical equations, pore-network modeling, and lattice-Boltzmann simulations, were applied to samples of sandy soils with distinct microstructural properties. The empirical equations showed varied results, highly dependent on the method chosen. The simulated sifting method was able to adequately estimate the granulometric properties of the soil, allowing the use of empirical permeability formulations for substantially small samples. Semi-theoretical equations based on the microstructural properties presented reasonable agreement for some samples. The pore-network modeling approach demonstrated computational efficiency but lacked accuracy. The lattice-Boltzmann method required significant computational resources but did not provide substantially closer alignment with the measured hydraulic properties of some samples. None of the simulations was able to properly determine the permeability of silty and organically contaminated sand. The study highlights the complexity of permeability estimation, emphasizing the need for choosing volumes of interest, resolution of micro-CT scans, and methods that match specific soil characteristics and available computational resources. An automated system for acquiring microscopic-resolution radiographic images of biological samples was developed. Mass-produced, low-cost, and easily automated components were used, such as Commercial-Off-The-Self CMOS image sensors (CIS), stepper motors, and control boards based on Arduino and RaspberryPi. System configuration, imaging protocols, and Image processing (filtering and stitching) were defined to obtain high-resolution images and for successful computational image reconstruction. Radiographic images were obtained for animal samples including the widely used animal models zebrafish (Danio rerio) and the fruit-fly (Drosophila melanogaster), as well as other small animal samples. The use of phosphotungstic acid (PTA) as a contrast agent was also studied. Radiographic images with resolutions of up to (7±0.6)μm were obtained, making this system comparable to commercial ones. This work constitutes a starting point for the development of more complex systems such as X-ray attenuation micro-tomography systems based on low-cost off-the-shelf technology. It will also bring the possibility to expand the studies that can be carried out with small animal models at many institutions (mostly those working on tight budgets), particularly those on the effects of ionizing radiation and absorption of heavy metal contaminants in animal tissues. Introduction: Exercise intensity determines the benefits of aerobic exercise. Our objectives were, in aerobic exercise at different intensities, to determine (1) changes in bone metabolism-related genes after acute exercise and (2) changes in bone mass, strength, remodeling, and bone formation-related proteins after long-term exercise. Materials and methods: Total 36 male C57BL/6J mice were divided into a control group and exercise groups at 3 different intensities: low, moderate, or high group. Each exercise group was assigned to acute- or long-term exercise groups. Tibias after acute exercise were evaluated by real-time PCR analysis. Furthermore, hindlimbs of long-term exercise were assessed by micro-CT, biomechanical, histological, and immunohistochemical analyses. Results: Acute moderate-intensity exercise decreased RANKL level as bone resorption marker, whereas low- and high-intensity exercise did not alter it. Additionally, only long-term exercise at moderate intensity increased bone mass and strength. Moderate-intensity exercise promoted osteoblast activity and suppressed osteoclast activity. After low- and high-intensity exercise, osteoblast and osteoclast activity were unchanged. An increase in the number of β-catenin-positive cells and a decrease in sclerostin-positive cells were observed in the only moderate group. Conclusion: These results showed that moderate-intensity exercise can inhibit bone resorption earlier, and long-term exercise can increase bone mass and strength through promoted bone formation via the Wnt/β-catenin activation. High-intensity exercise, traditionally considered better for bone, may fail to stimulate bone remodeling, leading to no change in bone mass and strength. This animal study was aimed to evaluate the efficacy of new bone formation and volume maintenance according to the particle type and the collagen membrane function for grafted octacalcium phosphate (OCP) in rabbit calvarial defects. The synthetic bone substitutes were prepared in powder form with 90% OCP and granular form with 76% OCP, respectively. The calvarial defects were divided into four groups according to the particle type and the membrane application. All specimens were acquired 2 weeks (n = 5) and 8 weeks (n = 5) after surgery. According to the micro-CT results, the new bone volume increased at 2 weeks in the 76% OCP groups compared to the 90% OCP groups, and the bone volume ratio was significantly lower in the 90% OCP group after 2 weeks. The histomorphometric analysis results indicated that the new bone area and its ratio in all experimental groups were increased at 8 weeks except for the group with 90% OCP without a membrane. Furthermore, the residual bone graft area and its ratio in the 90% OCP groups were decreased at 8 weeks. In conclusion, all types of OCP could be applied as biocompatible bone graft materials regardless of its density and membrane application. Neither the OCP concentration nor the membrane application had a significant effect on new bone formation in the defect area, but the higher the OCP concentration, the less graft volume maintenance was needed. Stress‐driven melt segregation may have important geochemical and geophysical effects but remains a poorly understood process. Few constraints exist on the permeability and distribution of melt in deformed partially molten rocks. Here, we characterize the 3D melt network and resulting permeability of an experimentally deformed partially molten rock containing several melt‐rich bands based on an X‐ray microtomography data set. Melt fractions range from 0.08 to 0.28 in the ∼20‐μm‐thick melt‐rich bands, and from 0.02 to 0.07 in the intervening ∼30‐μm‐thick regions. We simulated melt flow through subvolumes extracted from the reconstructed rock at five length scales ranging from the grain scale (3 μm) to the minimum length required to fully encompass two melt‐rich bands (64 μm). At grain scale, few subvolumes contain interconnected melt, and permeability is isotropic. As the length scale increases, more subvolumes contain melt that is interconnected parallel to the melt bands, but connectivity diminishes in the direction perpendicular to them. Even if melt is connected in all directions, permeability is lower perpendicular to the bands, in agreement with the elongation of melt pockets. Permeability parallel to the bands is proportional to melt fraction to the power of an exponent that increases from ∼2 to 5 with increasing length scale. The permeability in directions parallel to the bands is comparable to that for an isotropic partially molten rock. However, no flow is possible perpendicular to the bands over distances similar to the band spacing. Melt connectivity limits sample scale melt flow to the plane of the melt‐rich bands. Plain Language Summary: When a mixture of solid olivine grains and basaltic melt is subjected to differential stress, the melt can collect into sheet‐like bands. Melt transport properties, including melt trajectories, chemical diffusion, and ductile creep, may all become anisotropic in the presence of these melt‐rich bands. We present here the first quantification of the melt network properties in three dimensions based on synchrotron X‐ray microtomography of experimentally deformed olivine/basalt aggregates. We show that the melt network presents no connection between melt‐rich bands, restricting flow to the directions contained in the planes of these bands. Even at grain scale, melt pockets are preferentially oriented along the plane of the bands. The relationship between melt content and permeability derived from undeformed rocks is not valid at sample scale in sheared aggregates. This study utilises X-ray Computed Micro-Tomography (μXCT) as a non-destructive and non-invasive method to recover the original surface features and reveal the characteristics of encrusted, illegible Roman copper-based coins before any physical cleaning process is performed. The coins were retrieved from the topsoil during an archaeological survey in the countryside of the ancient city of Aquileia, Italy, and were severely degraded, covered with accumulated matter and pronounced encrustations developed over centuries of aging buried in soil. Despite their condition, most of the coins were identified from a numismatic standpoint using tomographic data alone, with the aid of reference images. They were subsequently cleaned using traditional manual methods and the results compared with μXCT. X-ray Fluorescence (XRF) analysis of the coins after the physical cleaning confirmed their numismatic attribution and revealed information regarding the influence of different alloy compositions on the applicability of the μXCT method, as well as on the corrosion process of the coins. This study showcases how the application of μXCT on fragile corroded metal artifacts prior to any intrusive manual procedures can expedite the identification process, mitigating the risk of information loss caused by physical handling and cleaning. This approach proves particularly valuable when dealing with large numbers of coins that would typically require restoration for identification purposes. It also emphasises the numerous advantages of using μXCT for coin identification, provenance determination, dating, virtual restoration, digitisation, and long-term preservation. The cultural heritage sector has increasingly explored the use of micro-CT (µCT) across numerous projects seeking to better understand past cultures and the materials they have left behind. As such, the role of micro-CT (µCT) is still being developed and projects continue to show novel ways that the technology can be adapted to. The Gjellestad ship, located in Halden (Østfold in Viken), Norway, is dated to the Viking Age and was found in a poor state of preservation. Both organic and metallic materials were deteriorated to the degree that standard excavation methods would have resulted in further damage to, or even the destruction of, these elements. A new approach was needed, and this presented an opportunity to explore the use for µCT as a documentative tool for field archaeology and conservation. As the remaining rivets were too fragile to handle directly, they were removed together with the surrounding matrix as soil blocks. To retain important stratigraphic and position information, a georeferencing system was developed that would be visible to µCT and included within each soil block. This enabled the spatial (re)positioning of the soil blocks by use of 3D GIS and in alongside with other spatial documentation gathered at the time of excavation. The quantity of soil blocks will give us a large dataset to work with and, although we continue to document the soil blocks with µCT, we now can discuss our preliminary results pertaining to the positive impact that µCT has for the documentation, conservation, and reconstruction of cultural heritage. Background: Breast microcalcifications (MCs) are considered to be a robust marker of breast cancer. A machine learning model can provide breast cancer diagnosis based on properties of individual MCs ‐ if their characteristics are captured at high resolution and in 3D. Purpose: The main purpose of the study was to explore the impact of image resolution (8 µm, 16 µm, 32 µm, 64 µm) when diagnosing breast cancer using radiomics features extracted from individual high resolution 3D micro‐CT MC images. Methods: Breast MCs extracted from 86 female patients were analyzed at four different spatial resolutions: 8 µm (original resolution) and 16 µm, 32 µm, 64 µm (simulated image resolutions). Radiomic features were extracted at each image resolution in an attempt, to find a compact feature signature allowing to distinguish benign and malignant MCs. Machine learning algorithms were used for classifying individual MCs and samples (i.e., patients). For sample diagnosis, a custom‐based thresholding approach was used to combine individual MC results into sample results. We conducted classification experiments when using (a) the same MCs visible in 8 µm, 16 µm, 32 µm, and 64 µm resolution; (b) the same MCs visible in 8 µm, 16 µm, and 32 µm resolution; (c) the same MCs visible in 8 µm and 16 µm resolution; (d) all MCs visible in 8 µm, 16 µm, 32 µm, and 64 µm resolution. Accuracy, sensitivity, specificity, AUC, and F1 score were computed for each experiment. Results: The individual MC results yielded an accuracy of 77.27%, AUC of 83.83%, F1 score of 77.25%, sensitivity of 80.86%, and specificity of 72.2% at 8 µm resolution. For the individual MC classifications we report for the F1 scores: a 2.29% drop when using 16 µm instead of 8 µm, a 4.01% drop when using 32 µm instead of 8 µm, a 10.69% drop when using 64 µm instead of 8 µm. The sample results yielded an accuracy and F1 score of 81.4%, sensitivity of 80.43%, and specificity value of 82.5% at 8 µm. For the sample classifications we report for F1 score values: a 6.3% drop when using 16 µm instead of 8 µm, a 4.91% drop when using 32 µm instead of 8 µm, and a 6.3% drop when using 64 µm instead of 8 µm. Conclusions: The highest classification results are obtained at the highest resolution (8 µm). If breast MCs characteristics could be visualized/captured in 3D at a higher resolution compared to what is used nowadays in digital mammograms (approximately 70 µm), breast cancer diagnosis will be improved. The aim of this study was to evaluate the remineralizing properties of ion-releasing restorative materials on pH cycling-induced carious dentin. Fifty sound molars were freshly extracted. The occlusal surfaces were abraded using water-cooled sandpaper (800 grit). The residual crowns were embedded in self-cured acrylic resin with the flat dentin surface exposed. A mesio-distal trench was created using a calibrated 0.5 mm deep occlusal reduction burr, and artificial dentin caries were generated by pH cycling. Then, teeth were randomly assigned to five groups according to the ion-releasing material used. For each sample, micro-CT acquisitions were performed at various intervals. Remineralization was assessed by mean gray value (MGV) measurements after registration and segmentation of the region of interest with 3D Slicer software. One-way repeated-measures ANOVA followed by Tukey's post hoc test was used to investigate the difference in MGVs among the various groups. Only Cention Forte showed significantly increased MGVs after 4 weeks compared to demineralized dentin. MGVs were higher, but not significantly, after placement of the restorative materials, including in the resin composite control group. These results can be explained by the radiopacity of the materials. Cention Forte, the material with the highest radiopacity, showed a significant increase in the MGVs of artificially carious dentin after 4 weeks. However, the study of dentin remineralization by micro-CT could be impacted by the radiopacity of the restorative materials used. This study focuses on characterizing a discontinuity within the Seán stalagmite (4.75–7.75 cm) by means of two nondestructive techniques: (1) high-resolution micro-computed tomography (micro-CT) and (2) X-ray fluorescence (XRF) core scanning (XRFCS). Micro-CT was used to study the stalagmite density, and XRFCS was applied to obtain the qualitative elemental composition and colour measurements. The new data obtained from nondestructive techniques have been combined with previously published geochemical data and fabric determinations from the same stalagmite found in Sa Balma des Quartó cave in Mallorca. The two methodologies applied in the present study have improved the characterization of the distinctive horizon. The micro-CT images identified the layer as a minor event due the high air content. The distinctive horizon is characterized by a high Ti-content, indicating the arrival of terrigenous particles. Based on those observations, together with the fact that the micrite layer appears filling the gaps between the older columnar fabric, we argue that the micrite layer may represent a major flooding event inside the cave after the year 1616 ± 23 CE and before the year 1623 ± 28 CE, which can be related to an extreme rainfall event. The application of biobased and biodegradable polymers, such as polylactide (PLA), in fused deposition modeling (FDM) 3D-printing technology creates a new prospect for rapid prototyping and other applications in the context of ecology. The popularity of the FDM method and its significance in material engineering not only creates new prospects for the development of technical sciences on an industrial scale, but also introduces new technologies into households. In this study, the kinetics of the hydrolytic degradation of samples obtained by the FDM method from commercially available PLA filaments under a thermally accelerated regime were analyzed. The investigation was conducted at the microstructural, supramolecular, and molecular levels by using methods such as micro-computed tomography (micro-CT), wide-angle X-ray diffraction (WAXD), viscosimetry, and mass erosion measurements. The obtained results clearly present the rapid structural changes in 3D-printed materials during degradation due to their amorphous initial structure. The complementary studies carried out at different scale levels allowed us to demonstrate the relationship between the observed structural changes in the samples and the hydrolytic decomposition of the polymer chains, which made it possible to scientifically understand the process and expand the knowledge on biodegradation. Objectives: The current clinical pulse lavage technique for flushing fresh osteochondral allografts (OCAs) to remove immunogenic elements from the subchondral bone is ineffective. This study aimed to identify the optimal method for removing immunogenic elements from OCAs. Methods: We examined five methods for the physical removal of immunogenic elements from OCAs from the femoral condyle of porcine knees. We distributed the OCAs randomly into the following seven groups: (1) control, (2) saline, (3) ultrasound, (4) vortex vibration (VV), (5) low‐pulse lavage (LPL), (6) high‐pulse lavage (HPL), and (7) high‐speed centrifugation (HSC). OCAs were evaluated using weight measurement, micro‐computed tomography (micro‐CT), macroscopic and histological evaluation, DNA quantification, and chondrocyte activity testing. Additionally, the subchondral bone was zoned to assess the bone marrow and nucleated cell contents. One‐way ANOVA and paired two‐tailed Student's t‐test are used for statistical analysis. Results: Histological evaluation and DNA quantification showed no significant reduction in marrow elements compared to the control group after the OCAs were treated with saline, ultrasound, or VV treatments; however, there was a significant reduction in marrow elements after LPL, HPL, and HSC treatments. Furthermore, HSC more effectively reduced the marrow elements of OCAs in the middle and deep zones compared with LPL (p < 0.0001) and HPL (p < 0.0001). Macroscopic evaluation revealed a significant reduction in blood, lipid, and marrow elements in the subchondral bone after HSC. Micro‐CT, histological analyses, and chondrocyte viability results showed that HSC did not damage the subchondral bone and cartilage; however, LPL and HPL may damage the subchondral bone. This study focuses on the mechanical properties of different thermoplastic polymers that have been processed by fused filament fabrication, including basic (PLA, PC, PA, ABS, PP, CPE, PET-G), industrial (PEI, PEKK), and with added functionality (thermochromic, electrostatic discharge, electrically conductive). Analysis of the porosity of specimens was performed by X-ray microtomography and optical microscopy of a fractured surface, both giving similar results. During printing, a non-equilibrium polymer macromolecular structure was created. The stiffness and strength of the printed specimens were impacted by the stabilization of the macromolecular structure over time. The maximum values were reached for amorphous materials after 24 hours and for semi-crystalline materials after 160 hours. Tensile properties of "as-received" filaments, extruded mono-fibers, and unidirectional printed specimens were compared. In most cases, the elastic modulus of "as-received" filaments was lower than that of the extruded mono-fibers by 12% on average. Loading rate significantly affects both elastic modulus and strength, confirming the essential contribution of the viscoelastic component to the whole deformability of polymers. The elastic modulus and strength increased by 20 and 80%, respectively. The effects of layer thickness and nozzle diameter on mechanical properties were investigated. The compatibility of different polymer types for hybrid structures was evaluated in the adhesion tests. Tests showed that adhesion at hybrid PLA contact is only slightly affected by the presence of colourant additives in one of the parts. The amalgamation of veterinary anatomy, technology and innovation has led to development of latest technological advancement in the field of veterinary medicine, i.e., three-dimensional (3D) imaging and reconstruction. 3D visualization technique followed by 3D reconstruction has been proven to enhance non-destructive 3D visualization grossly or microscopically, e.g., skeletal muscle, smooth muscle, ligaments, cartilage, connective tissue, blood vessels, nerves, lymph nodes, and glands. The core aim of this manuscript is to document non-invasive 3D visualization methods being adopted currently in veterinary anatomy to reveal underlying morphology and to reconstruct them by 3D softwares followed by printing, its applications, current challenges, trends and future opportunities. 3D visualization methods such as MRI, CT scans and micro-CT scans are utilised in revealing volumetric data and underlying morphology at microscopic levels as well. This will pave a way to transform and re-invent the future of teaching in veterinary medicine, in clinical cases as well as in exploring wildlife anatomy. This review provides novel insights into 3D visualization and printing as it is the future of veterinary anatomy, thus making it spread to become the plethora of opportunities for whole veterinary science. Background: It is now well established that physical exercise is an effective preventive method to reduce and treat certain chronic diseases, particularly musculoskeletal disorders. At the bone level, running exercise is well known for its positive effects on various parameters of bone quality. There is, however, no consensus regarding the effects of different running exercise modalities on bone quality. Aim: The objective of this study was to compare the effects of three treadmill running modalities: intermittent, moderate continuous, and a combination of both—on bone quality parameters in rats. Methods: Thirty-nine, 5-week-old, male Wistar rats were randomly divided in 4 groups: sedentary control (SED; n = 10), intermittent running exercise (IE; n = 10), continuous running exercise (CE; n = 10) and combined running exercise (COME; n = 9). Rats in running groups were exercised 45 min/day, 5 days/week, for 8 consecutive weeks. Femoral micro-architectural parameters were assessed by micro-CT; femoral osteocyte apoptosis, osteoclast resorption and bone histomorphometry were assessed by histology. Results: Femoral trabecular thickness in the combined running group was increased (p < 0.0001) compared to respective results in the other running groups (0.13 mm vs 0.11 mm). The cortical thickness, osteocyte lacunae occupancy rate in the whole femur, numbers of apoptotic osteocytes and osteoclastic resorption surfaces were not significantly different between groups. Statistical differences were occasionally noted depending on the femoral anatomical region. Porous geopolymers with sustainable dimensional stability at high temperatures represent a noteworthy challenge for the development of thermomechanical, reliable and multifunctional composites. This study reports on the porosity formation of refractory-reinforced geopolymer paste via hydrogen peroxide decomposition. Reinforcing the slurry with ceramic chamotte and cordierite increases the apparent viscosity more than eight times in the interval to 100 s−1 retaining a pseudoplastic behavior. Cordierite particles increase the plastic viscosity at 6 Pa s and yield stress above 28 Pa. Surprisingly, the particulate ceramic filler acts as a porosity-modifier thereby reducing the mesoporosity region. Microstructural and porosity behavior was studied via digital microscopy, SEM, MIP and micro-CT. The wrinkled morphology of ceramics significantly affects the macroporosity formation in the region of 100 μm reaching a total porosity of ∼60 vol%. The presence of large pores in the macro region and the high distribution of interconnected porosity in ceramics-reinforced composite foams resulted in compressive strengths ranging from 2 to 10 MPa. The effect of curing rate on porosity was most evident in the cordierite reinforcement which led to the occurrence of large interconnected pores. Based on thermomechanical analysis, the ceramic-reinforced foams show high dimensional stability with a reduction in total shrinkage to −0.71 % at 850 °C. Coal is a natural and complex porous medium, which contains pore-fracture structures of different scales. The pore-fracture structures of coal directly affect the characteristics of gas storage and migration. In this paper, high-resolution X-ray CT scanning technology is used to quantitatively characterize the pore-fracture structure of medium and high-rank coals. First, the porosity inversion method is used to determine the optimal threshold segmentation of pores and fracture in medium and high-rank coals. Then, the pore and fracture model of representative element volume (REV) was extracted and established to characterize the area porosity, pore surface area, and shape factor of the two coals, and the relationship between pore surface area and shape factor was investigated. Finally, an equivalent pore network model (PNM) of connected pore topology was established, and pore-throat parameters are statistically analyzed, including pore volume, pore radius, throat radius, throat length, and coordination number. The results show that the average pore diameter and average surface area of macropores in medium-rank coals are larger than those of high-rank coals, and high-rank coals have fewer isolated pores. With the increase of coal rank, the length and radius of the throat gradually decrease, which indicates that the throat of the medium-rank coal has a higher degree of development, and the gas seepage ability in the throat is stronger. The effect of CVI-induced porosity on mechanical behaviour of C f /SiC composites possessing 2.5D (woven) and 3D (NOOBed) architectures with multilayered (PyC/SiC) n=4 interphase has been investigated. X-ray computed micro-tomography has shown that inter-bundle and inter-fibre pore distribution is influenced by fibre architecture. Elastic constants measured by ultrasound phase spectroscopy have revealed greater anisotropy in 2.5D C f /SiC composite than 3D C f /SiC composite. Owing to negligible C f fraction and flattened inter-bundle pores distributed in thickness direction of 2.5D C f /SiC composite, the elastic constant, C 11 (∼15±0.03 GPa) is <1/6th of C 22 and C 33 obtained along fibre directions. Despite the flexural strength of 2.5D C f /SiC composite (∼335±8 MPa) being ∼24% higher than 3D C f /SiC composite, a more equitable distribution of C f along three mutually orthogonal directions leads to higher fracture toughness of 3D C f /SiC composites (∼19±1 MPa-m1/2) and greater damage tolerance. Purpose: Previous studies have shown that DNA methyltransferase 3b (Dnmt3b) is the only Dnmt responsive to fracture repair and Dnmt3b ablation in Prx1-positive stem cells and chondrocyte cells both delayed fracture repair. Our study aims to explore the influence of Dnmt3b ablation in Gli1-positive stem cells in fracture healing mice and the underlying mechanism. Methods: We generated Gli1-CreERT2; Dnmt3bflox/flox (Dnmt3bGli1ER) mice to operated tibia fracture. Fracture callus tissues of Dnmt3bGli1ER mice and control mice were collected and analyzed by X-ray, micro-CT, biomechanical testing, histopathology and TUNEL assay. Results: The cartilaginous callus significantly decrease in ablation of Dnmt3b in Gli1-positive stem cells during fracture repair. The chondrogenic and osteogenic indicators (Sox9 and Runx2) in the fracture healing tissues in Dnmt3bGli1ER mice much less than control mice. Dnmt3bGli1ER mice led to delayed bone callus remodeling and decreased biomechanical properties of the newly formed bone during fracture repair. Both the expressions of Caspase-3 and Caspase-8 were upregulated in Dnmt3bGli1ER mice as well as the expressions of BCL-2. Conclusions: Our study provides an evidence that Dnmt3b ablation Gli1-positive stem cells can affect fracture healing and lead to poor fracture healing by regulating apoptosis to decrease chondrocyte hypertrophic maturation. Guided Bone Regeneration is a common procedure, yet, as new grafting materials are being introduced into the market, a reliable evaluation method is required. Critical size defect in animal models provides an accurate simulation, followed by histological sections to evaluate the new bone formation. However, histology is destructive, two-dimensional and technique-sensitive. In this study we developed a novel volumetric Micro-CT analysis to quantify new bone formation characteristics. Eight adult female New Zealand white rabbits were subjected to calvarial critical-size defects. Four 8 mm in diameter circular defects were preformed in each animal, to allow random allocation of four treatment modalities. All calvarias were scanned using Micro-CT. Each defect was segmented into four equal parts: pristine bone, outer, middle, and inner. Amira software (v. 6.3, www.fei.com) was used to calculate the new bone volume in each region and compare it to that of the pristine bone. All grafting materials demonstrated that new bone formation decreased as it moved inward. Only the inner region differed across grafting materials (p = 0.001). The new Micro-CT analysis allowed us to divide each defect into 3D regions providing better understanding of the bone formation process. Amongst the various advantages of the Micro-CT, it enables us to quantify the graft materials and the newly formed bone independently, and to describe the defect morphology in 3D (bi- vs. uni-cortical defects). Providing an insight into the inner region of the defect can better predict the regenerative potential of the bone augmentation graft material. Objectives: The purpose of this study was to investigate the imaging characteristics of medication-related osteonecrosis of the jaw (MRONJ) using [18F]fluoride positron emission tomography/computed tomography (PET/CT) and [18F]fluorodeoxyglucose (FDG) PET/magnetic resonance imaging (MRI) for preoperative assessment and to correlate them with microarchitectural and histomorphometric data with respect to clinical findings. Methods: Twelve patients (five female; mean age 75 ± 7.6 yr) with symptomatic MRONJ underwent both scans on the same day, and imaging findings were used to plan surgical interventions for seven patients. Bone tracer uptake was classified as high, medium, or low, and surgical samples were evaluated using Micro-CT and histomorphometric analysis. Results: CT showed medullary sclerosis in all patients, and MRI revealed gadolinium enhancement in four patients. PET imaging revealed remarkably elevated [18F]fluoride uptake and moderately increased [18F]FDG uptake in MRONJ compared to healthy jawbones, with both differences being statistically significant. [18F]fluoride uptake was associated with necrosis, bacteria, and inflammatory tissue. Micro-CT data did not show significant differences, but histomorphometric analysis revealed higher osteocyte and lacunae densities in the high [18F]fluoride uptake group, and more necrotic bone in the medium [18F]fluoride uptake group. Bacteria were observed in all areas. Conclusions: In summary, [18F]fluoride PET accurately identified MRONJ extent, revealing functional changes in jawbone remodeling not visible on CT. [18F]FDG PET showed differences in bone and soft tissue, though less pronounced. The physical properties of Ti6Al4V powder affect the spreadability of the powder and uniformity of the powder bed, which had a great impact on the performance of built parts made by powder bed fusion technology. Micro-computed tomography is a well-established technique used to analyze the non-destructivity of the objects' interior. Ti6Al4V powders were scanned with micro-CT to show the internal and external information of all the particles. The morphology, particle size distribution, hollow particle ratio, density, inclusion, and specific surface area of the powder samples were quantitatively characterized, and the relationship of flowability with these physical properties was analyzed in this work. The research results of this article showed that micro-CT is an effective way to characterize these items, and can be developed as a standard method of powder physical properties in the future. In this study, we compared the material properties of linearly and sharply graded Ti6Al4V additively manufactured samples to investigate whether the more severe discontinuities caused by sharp grading can reduce performance. We performed compression testing with digital image correlation (DIC) in two loading directions for each grading design to simulate iso-stress and iso-strain conditions. We extracted the elastic stiffness, yield strength, yield strain, and energy absorption capacity of each sample. In addition, we used micro-computed tomography (micro-CT) imaging to examine the printing quality and dimensional accuracy. We found that sharply graded struts have a 12.95% increase in strut cross-sectional areas, whereas linearly graded struts produced an average of 49.24% increase compared to design. However, sharply graded and linearly graded FGL samples do not have statistically significant differences in elastic stiffness and yield strength. For the iso-strain condition, the average DIC-corrected stiffnesses for linearly and sharply graded samples were 6.15 GPa and 5.43 GPa, respectively (p = 0.4466), and the yield stresses were 290.4 MPa and 291.2 MPa, respectively (p = 0.5734). Furthermore, we confirmed different types of printing defects using micro-CT, including defective pores and disconnected struts. These results suggest that the loss of material properties caused by manufacturing defects outweighs the adverse effects of discrete-grading-induced discontinuities. [ABSTRACT FROM AUTHOR]

In many taxa, females store sperm in specialized storage organs. Most insect sperm storage organs have a tubular structure, typically consisting of a central lumen surrounded by epithelial cells. These specialized tubules perform the essential tasks of transporting sperm through the female reproductive tract and supporting long-term sperm survival and function. Little is known about the way in which female sperm storage organs provide an environment conducive to sperm survival. We address this using a combined light microscopy, micro computed tomography (microCT), and Focused Ion Beam Scanning Electron Microscopy (FIB-SEM) approach for high-resolution correlative three-dimensional imaging to advance our understanding of sperm-female interactions in Drosophila melanogaster. Using this multimodal approach, we were able to scan the lower female reproductive tract and distal portion of the seminal receptacle at low magnification, and to subsequently zoom in for further analysis on an ultrastructural level. Our findings highlight aspects of the way in which the seminal receptacle keeps sperm viable in the lumen, and set the stage for further studies. The methods developed are suitable not only for Drosophila but also for other organisms with soft, delicate tissues. The characterization of the vibrations of the middle ear ossicles during sound transmission is a focal point in clinical research. However, the small size of the structures, their micrometer-scale movement, and the deep-seated position of the middle ear within the temporal bone make these types of measurements extremely challenging. In this work, dynamic synchrotron-based X-ray phase-contrast microtomography is used on acoustically stimulated intact human ears, allowing for the three-dimensional visualization of entire human eardrums and ossicular chains in motion. A post-gating algorithm is used to temporally resolve the fast micromotions at 128 Hz, coupled with a high-throughput pipeline to process the large tomographic datasets. Seven ex-vivo fresh-frozen human temporal bones in healthy conditions are studied, and the rigid body motions of the ossicles are quantitatively delineated. Clinically relevant regions of the ossicular chain are tracked in 3D, and the amplitudes of their displacement are computed for two acoustic stimuli. Synchrotron X-ray imaging of stimulated human middle ears reveals the 3D movement of auditory ossicles: it enables their motion quantification, confirms the lever-like motions of the malleus and incus and reveals a more intricate stapedial behavior. Type 2 diabetes interferes with bone remodeling mechanisms, requiring studies to reverse this damage, and resveratrol is a polyphenol with rich properties. This study aimed to characterize the long bone morphology and peri-implant biomechanics of normoglycemic and type 2 diabetic animals treated with resveratrol. Thirty-two male Wistar rats were used and divided into normoglycemic and diabetic with or without treatment. They had the installation of implants in the tibia and treatment with oral resveratrol within 45 days. Resveratrol was responsible for weight homeostasis and decreased glycemic levels in rats with type 2 diabetes. The three-point bending testing, resveratrol showed positive effects on the biomechanics of long bones, corroborating a more resistant bone in comparison to untreated diabetics. Micro-ct revealed how bone metabolism is affected by systemic disease, decreasing bone quality. The counter-torque of normoglycemic animals showed superior osseointegration to diabetes, with no differences in the administration of the polyphenol, showing the sovereignty of the deleterious effects of the disease when there is a tissue lesion and an inflammatory picture installed. Overall, resveratrol acted positively in the etiopathogenesis of type 2 diabetes and revealed positive effects on the strength of long bones. [ABSTRACT FROM AUTHOR]

Archaeocyaths are a group of extinct filter feeders that flourished in the early Cambrian period and occupied an important position in the evolution of basal fauna and the early marine ecosystem. However, the detailed morphological and anatomical information of this group are still unclear due to insufficient fossil material and limited experimental analyses. Here, we report exquisitely preserved phosphatized archaeocyathan fossil cups, ca. 515 million years old, from the top of the Shuijingtuo Formation (Series 2, Stage 3) and the Xiannüdong Formation (Series 2, Stage 3) of the Yangtze Platform, South China. Detailed observation of their external morphology via scanning electron microscopy (SEM) and micro-computed tomography (Micro-CT) analysis revealed detailed information of their internal structure. They have a typical double-walled cup, with the perforated inner and outer walls concentrically distributed, but the structure between the two walls differs. The inverted cone-shaped cups have radially distributed septa between the walls. Perforated septa connect the two walls. The low and columnar cups have canals between the two walls, forming the network. These pores and cavities constitute an important component of the water current system (pumping and filtering water with a network of canals and chambers) and influence the process of filtration in the cup. In comparison to traditional thin-section analysis, the combination of SEM and Micro-CT analysis on phosphatized archaeocyaths presented in this study further explored the detailed internal structure and finely reconstructed the microscopic overall morphology and anatomy, which provide important information to help us understand the systematic taxonomy, anatomy, and morphology of archaeocyaths during the Cambrian period.
